La paziente non deve essere in periodo mestruale, non deve aver urinato nelle ultime 3 ore e non deve eseguire lavande vaginali prima del prelievo. Inoltre deve astenersi da rapporti sessuali nelle 24 ore precedenti l’esame. La raccolta del campione è ottenuta con l’aiuto di uno speculum. Dopo aver rimosso il tappo mucoso cervicale con un batuffolo di cotone, si introduce un tampone sterile nel canale cervicale e con movimento rotatorio si raccoglie sia l’essudato che le cellule cervicali.
